Identification and Characterization of Two Putative Citrus Phosphomannose Isomerase (CsPMI) Genes as Selectable Markers for Mature Citrus Transformation

Two Citrus sinensis (L.) phosphomannose isomerase (PMI) genes, CsPMI1 and CsPMI2, were evaluated as novel selectable markers in mature citrus transformation. Transgenic shoots produced after transformation of Kuharske rootstock with each PMI construct selected on six treatments mannose sucrose. For CsPMI1, there no significant differences among the various sucrose for mean number positive (PS), efficiency based (TES), or explants (TEE). However, CsPMI2 gene, transgenics two (7.5 g L−1 + 22.5 15 sucrose) was significantly greater than control TES at 4.2% 3.7%, respectively. Moreover, TEE treatment, supported value. Most transgenic lines demonstrated higher vivo vitro enzyme assays compared wild-type control. provided acceptable selection citrus, it will be applied future intragenic research.
